Radiation (RT) and androgen deprivation (ADT) are important modalities for the treatment of patients diagnosed with early localized and advanced metastatic PCa respectively. These approaches (i) ultimately lead to resistance and recurrent cancer and (ii) are associated with side effects that impact quality of life significantly. We propose that “downregulation of ribosomal protein S6 (p70S6K encoded by RPS6KB1)” will be effective in preventing recurrence. This premise is based on our observations that (i) human prostate tumors show significantly elevated levels of p70S6K and (ii) genetic and pharmacologic inhibition of RPS6KB1 enhances the effectiveness of radiation in PCa cells.
